GATA-3 is a zinc finger transcription factor and plays an important role in promoting and directing cell proliferation, development, and differentiation in many tissues and cell types. GATA-3 expression is primarily seen in breast and urothelial carcinomas. 'Therefore, GATA3 antibody can be useful in the identification of unknown primary carcinoma when carcinomas of the breast or bladder are a possibility. The metal-responsive element (MRE)-binding transcription factor (MTF-1) stimulates the expression of metallothioneins in response to the exposure of cells to heavy metals. MTF-1 contains six zinc fingers in the DNA binding domain. The phosphorylation of MTF-1 in response to metal exposure appears to play a significant role in the ability of MTF-1 to activate metallothionein transcription. In addition to its role in metallothionein activation, MTF-1 is involved in a post-transcription regulatory complex for ribosomal protein S25. MTF-1, La and p53 inhibit the nuclear export of S25 mRNA in response to nutrient depravation. Furthermore, MTF-1 acts as a chromatin insulator on integrated transgenes in cultured cells to insulate active loci against chromatin silencing. AKT1 is a 62 kDa protein and member of the serine/threonine kinase AKT family. AKT1 is also known as Protein Kinase B (PKB) or Rac Protein Kinase Alpha. The Akt family also includes AKT2. Activation of AKT1 by insulin or insulin-growth factor-1 (IGF-1) results in phosphorylation of both Thr 308 and Ser 473. AKT proteins become phosphorylated and activated in insulin/IGF-1-stimulated cells by an upstream kinase(s), and the activation of AKT1 and AKT2 is inhibited by the PI kinase inhibitor wortmannin. In the prostate the protein is up-regulated in cancerous cells and is used as an effective diagnostic and prognostic indicator of prostate cancer. Ki-67 antigen is a nuclear, non-histone protein that is present in all stages of the cell cycle except G0. This characteristic makes Ki-67 an excellent marker for proliferating cells and is commonly used as one of the prognostic factors in cancer studies. A correlation has been demonstrated between Ki-67 index and the histo-pathological grade of neoplasms. Assessment of Ki-67 expression in renal and ureter tumors shows a correlation between tumor proliferation and disease progression, thus making it possible to differentiate high-risk patients. Ki-67 expression may also prove to be important for distinguishing between malignant and benign peripheral nerve sheath tumors. Ki-67 labeling index has been shown to be a prognostic marker in a number of neoplasms including grade II astrocytoma, oligodendroglioma, colon carcinoma, and breast carcinoma. EBAG9, also known as RCAS1, is an estrogen-transcribed protein. Soluble and membranous RCAS1 proteins may play a role in the immune escape of tumor cells by promoting T lymphocyte inhibition of growth and apoptosis. RCAS1 is expressed in a wide variety of cancers, including uterine, ovarian, and lung cancer cells, and acts as a ligand for a putative receptor present on peripheral lymphocytes. RCAS1 is highly expressed not only in cancer cells but also in non-tumor bile duct cells subject to immune attack. RCAS1 inhibits the in vitro growth of receptor-expressing cells and induces apoptosis, contributing to the ability of tumor cells to evade host immune surveillance.
